I'm going to answer this in typical mailing list fasion.  "It depends".
Basically, if you modify your Makefile for something that hasn't been
completed yet, then simply re-runing it will certainly do the trick.
However, if you change a key file(s), such as uClibc itself, then you
need to clear out anything that was compiled against it and run the
process again.
